On April 14, 2001, Madison Square Garden in New York City hosted a significant middleweight unification bout between Bernard Hopkins and Keith Holmes. This fight marked the opening round of the 2001 Middleweight Unification Tournament, organized by promoter Don King to crown the first undisputed middleweight champion since Marvin Hagler in 1987.

Hopkins entered the ring as the IBF Middleweight Champion, making his 13th title defence, while Holmes held the WBC Middleweight Title and was defending for the third time. The bout was televised on HBO World Championship Boxing, with Jimmy Lennon Jr. serving as the ring announcer.

The lead-up to the fight was characterized by bold predictions and verbal sparring. Hopkins confidently declared he would knock out Holmes, stating he didn't deserve to win if the fight went the distance. For his part, Holmes questioned Hopkins' credentials and accused him of fighting dirty.

At the weigh-in, Hopkins came in at 159 pounds, while Holmes was slightly lighter at 157 pounds. The fight saw Hopkins dominate from the third round onward, employing an effective body attack and occasionally troubling Holmes with headshots. However, controversy arose over low blows, with Hopkins losing a point in the fifth round and two timeouts being called for Holmes due to low blows. Hopkins attributed this to Holmes wearing unusually high trunks.

CompuBox statistics highlighted Hopkins' control of the fight, as he consistently outworked and outlanded Holmes throughout the 12 rounds. The judges' scorecards reflected this dominance: Tom Kaczmarek scored it 119-108, George Colon had it 118-109, and Larry Hazzard Jr. saw it 117-110, all favouring Hopkins. HBO's unofficial judge, Harold Lederman, also had Hopkins winning convincingly with a score of 109-99 after 11 rounds.

Post-fight, Hopkins acknowledged his failure to deliver on his knockout prediction but expressed satisfaction with his performance. Holmes, showing sportsmanship, embraced Hopkins and admitted that his long layoff might have affected his performance.

This victory propelled Hopkins to the final of the Middleweight Unification Tournament, setting up a showdown with Félix Trinidad, who had defeated William Joppy for the WBA belt a month later. The tournament aimed to produce the first undisputed middleweight champion in over a decade, adding historical significance to Hopkins' win over Holmes.
